What is SS-31, and what is it studied for?
SS-31, known in pharmacology as elamipretide, is the tetrapeptide D-Arg-Dmt-Lys-Phe-NH2, formula C32H49N9O5, mass 639.8. Its alternating aromatic and cationic residues concentrate it in the inner mitochondrial membrane, where it binds cardiolipin, and it is studied in cell and animal models of mitochondrial bioenergetics. A pharmaceutical form received FDA accelerated approval in September 2025 under the name Forzinity; the material here is for in-vitro research only.
What is the structure of SS-31?
Four residues: a D-arginine, 2,6-dimethyltyrosine, lysine and phenylalanine, C-amidated. The D-configuration and the non-natural dimethyltyrosine are what make it resistant to peptidases; neither is visible to mass spectrometry, so the certificate confirms mass and composition, not chirality. Two cationic residues give a net positive charge that drives mitochondrial accumulation.

How is SS-31 thought to work?
Binds cardiolipin in the inner mitochondrial membrane and is reported to stabilise cristae structure and electron-transport efficiency, reducing electron leak; Birk and colleagues described the cardiolipin interaction in 2013 and Szeto reviewed the pharmacology in 2014. It is not a free-radical scavenger in the usual sense; its described action is structural.
