What is pinealon, and what is it studied for?
Pinealon is the tripeptide Glu-Asp-Arg, formula C15H26N6O8, mass 418.4, one of the short synthetic peptides designed by the St Petersburg bioregulator programme to reproduce the composition of a pineal extract. It is studied in cultured cells and rodents, almost entirely by the group that designed it, for effects on cell viability and free-radical measures. No human trial is registered.
What is the structure of Pinealon?
Three residues, glutamic acid, aspartic acid and arginine, written EDR, with free termini and no modifications. At neutral pH the two side-chain carboxyls, the C-terminal carboxyl, the arginine guanidinium and the N-terminal amine make it a small, highly polar zwitterion with a net charge near minus one, which is why it dissolves instantly and elutes early on a reversed-phase column.

How is Pinealon thought to work?
The designers propose that these short peptides enter cells and interact with DNA directly, altering transcription; the proposal is argued in their papers and is not an established receptor pharmacology. No receptor for pinealon has been identified. In the cell studies that exist, the measured outputs are viability and markers of oxidative stress, read after exposure of cultured cells to the peptide.

What does the published literature show?
Khavinson and colleagues reported in Rejuvenation Research in 2011 that pinealon raised viability and lowered free-radical levels in cultured cells. Anisimov and Khavinson's 2010 Biogerontology review places it in the pineal series and summarises rodent work. Independent replication is sparse and there is no registered clinical study; the material here is for in-vitro and preclinical research only.
How is Pinealon handled in the laboratory?
Dissolves in water or buffer immediately. No cysteine, methionine or tryptophan, so the usual oxidation routes do not apply; aspartate can slowly isomerise at acidic pH, so hold stocks near neutral. Is pinealon the same as epithalon?
No. Pinealon is Glu-Asp-Arg, three residues, mass 418.4. Epithalon is Ala-Glu-Asp-Gly, four residues, mass 390.3. Both come from the pineal programme; the sequences and masses differ.

Why is a tripeptide sold in a 20 mg vial?
Because molar amount is what an experiment consumes. 20 mg of a 418-dalton peptide is 47.8 micromoles, far more than a 5 mg vial of a long peptide holds.
